Descrição detalhada : The Leucine-rich repeat protein SHOC-2 (SHOC2) is the regulatory subunit of protein phosphatase 1 (PP1c) that acts as a M-Ras/MRAS effector and plays a key role in MAPK pathway activation. Upon RAS activation, SHOC2 acts as a positive modulator of the RAS-MAPK signaling via specifically allowing PP1c to dephosphorylate the 'Ser-259' inhibitory site of RAF1 kinase and stimulate RAF1 activity. SHOC2 localizes to cytoplasm when inactive but rapidly translocates to the nucleus when activated, such as during growth factor stimulation. Thus, as expected, SHOC2 also participates in FGF receptor signaling. Molecularly, SHOC2 is a scaffold protein that brings together activated Ras (activated via G-protein coupled receptors and the interaction of GTP to GDP) and Raf, which then facilitates the transfer of the phospho-Raf phosphate to other proteins for the activation of the MAP Kinase pathway for gene activation. Defects in SHOC2 lead to a type of multi-symptomatic disorder termed the Noonan Syndrome Loss of Anagen Hair (NSLAH) that affects everything from skeletal formation to facial features and many organs including the brain, skin, muscles, and heart.
