Descrição detalhada : Serine/threonine-protein kinase Nek6 (UniProt: Q9HC98; also known as EC: 2.7.11.1, Never in mitosis A-related kinase 6, NimA-related protein kinase 6, Protein kinase SID6-1512) is encoded by the NEK6 gene (Gene ID: 10783) in human. Nek6, a member of the NIMA-related serine/threonine kinase family that plays an important role in mitotic cell cycle progression and is required for chromosome segregation at metaphase-anaphase transition. It localizes to spindle microtubules in metaphase and anaphase and to the mid-body during cytokinesis. Nek6 is ubiquitously expressed, with highest expression in heart and skeletal muscle. Its levels are up-regulated in a variety of malignant cancers, such as breast, colon, lung, and gastric cancers. Nek6 is shown to be directly phosphorylated by Chk1 and Chk2, suggesting that it is a direct target of the DNA damage checkpoint. It is also phosphorylated upon IR or UV-induced DNA damage. Nek6 is activated by phosphorylation at Ser206. Although expression of Nek6 is tightly controlled in normal cells, this protein is highly overexpressed in various human cancers and regulates tumor progression and its expression and activity levels directly correlate to tumorigenicity of human cancer cells.
