Descrição detalhada : Endoplasmic reticulum (ER) mannosyl-oligosaccharide 1,2-alpha-mannosidase (EC 3.2.1.113; UniProt Q9UKM7), also known as ER mannosyl-oligosaccharide 1,2-alpha-mannosidase, ER alpha-1,2-mannosidase, ERMan1, ERManI, ER mannosidase 1, Man9GlcNAc2-specific-processing alpha-mannosidase, Mannosidase alpha class 1B member 1) is encoded by the MAN1B1 gene in human. Originally thought to function in a similar fashion as its yeast ortholog Mns1p, which removes a terminal mannose unit to initiate a glycan-based ER-associated degradation (ERAD) signal. Mammalian MAN1B1 protein is actually localized in Golgi instead of ER. MAN1B1 protein participates in a Golgi-based quality control network that captures misfolded protein substrate, which escape from ER, most likely via COPII vesicles, and recycles them back to the ER for ERAD. Upon entering the Golgi, the initially escasped unfold proteins are retained by a cargo-capturing complex in which non-enzymatic regions of MAN1B1, including the decapeptide sequence (DPS) within the MAN1B1 luminal stem, play an integral role. Facilitated by direct interactions with gamma-COP, MAN1B1 promotes the recruitment of unfold proteins into COPI vesicles, which then recycles the unfold proteins back to the ER via retrograde COPI vesicles without the need for trimming by MAN1B1 protein.
