Descrição detalhada : Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone H3 variants (H3.1, H3.2 and H3.3) have been implicated in the epigenetic memory of cellular state. Genome-wide patterns of H3 are dependent on amino acid sequence and change with cellular differentiation at developmentally regulated loci. Histone H3 is reported to be acetylated at Lys64 (H3K64ac) by p300 co-activator. In addition, H3K64ac is found enriched at the transcriptional start sites of active genes, consistent with a transcriptional activation modification opposite of its repressive counterpart H3K64me3 (PMID 24668167). The target specificity of ABE1057 is confirmed with peptide-based epitope analysis by dot blot, using Cat. No. 16-667, AbSurance Histone H3 Antibody Specificity Array, and Cat. No. 16-665, AbSurance Histone H2A, H2B, H4 Antibody Specificity Array. Sequence surrounding the target acetylation site is conserved among human Histone H3 subtypes H3.1, H3.2, H3.3, and H3.3C/H3.5 (UniProt P68431, Q71DI3, P84243, and Q6NXT2, respectively).
