Descrição detalhada : Tyrosine-protein kinase ITK/TSK (UniProt: Q08881; also known as EC:2.7.10.2, Interleukin-2-inducible T-cell kinase, IL-2-inducible T-cell kinase, Kinase EMT, T-cell-specific kinase, Tyrosine-protein kinase Lyk) is encoded by the ITK (also known as EMT, LYK) gene (Gene ID: 3702) in human. ITK/TSK is a protein tyrosine kinase that plays an essential role in regulation of the adaptive immune response. It regulates the development, function, and differentiation of conventional T-cells and non-conventional natural killer T-cells. It contains four zinc binding sites. It s N-terminal PH domain allows it to be recruited to the plasma membrane by an activated PI3 kinase. This domain also contains a proline-rich region (PRR). The adjoining SH3 domain binds to PRR. The SH2 domain is required for binding tyrosine-phosphorylated substrates and the kinase domain located in the C-terminal region is required for tyrosine phosphorylation. ITK/TSK is phosphorylated at Tyr512 in the activation loop of the kinase domain by LCK and subsequent autophosphorylation at Tyr180, which lies within the substrate-binding sequence in the SH3 domain, allows the kinase activation. Mutations in ITK gene are reported to cause lymphoproliferative syndrome 1, a rare immunodeficiency characterized by extreme susceptibility to infection with Epstein-Barr virus that can lead to fatal outcome.
