Descrição detalhada : Liver receptor homolog 1 (LRH-1, CPF, FTF, PHR-1, FTF, or hB1F) is an orphan nuclear receptor that is highly expressed in the intestines, pancreas, ovary, and liver. LRH-1 is a constitutively active transcription factor with a complex role in many cellular processes which may include lipid metabolism, cell proliferation, development, reproduction, and immunity. LRH-1 may interact with an array of transcriptional coactivators that modulate its activity, among which are the MBF1, SRC, and PGC-1alpha proteins. In addition, ERK phosphorylation on Ser-238 and Ser-243 also enhances LRH-1 activity. However, LRH-1 is also negatively regulated by several factors. For instance, corepressors such as SHP, DAX-1, and PROX-1, preclude its binding to DNA; and sumoylation promotes its inactivation and sequestration into nuclear bodies. LRH-1 targets many genes including the CYP7A1 and CYP8B1 genes involved in bile synthesis; and Oct4 required for pluripotency in embryonic stem cells. LRH-1 has been implicated in inflammatory bowel disease, and tumor development in the intestines and pancreas.
