Descrição detalhada : SHC-transforming protein 1 (UniProt P29353; also known as SH2 domain protein C1, SHC-transforming protein 3, SHC-transforming protein A, Src homology 2 domain-containing-transforming protein C1) is encoded by the SHC1 (also known as SHC, SHCA) gene (Gene ID 6464) in human. Originally identified as an SH2-containing proto-oncogene involved in growth factor signaling, Shc proteins function as adaptor molecules recruited by numerous receptors (e.g. integrins, GPCRs, and receptors for growth factors, antigens, cytokines, and hormones) for signling. In addition to the ubiquitously expressed Shc (a.k.a. ShcA by SHC1/SHCA gene), there exist also ShcB (by SHC2/SHCB gene) and ShcC (by SHC3/SHCC gene), which are predominantly expressed in neuronal cells. Alternative splicings result in six ShcA isoforms, including p66Shc, p52Shc, and p46Shc. All ShcA isoforms possess two distinct domains, PTB (a.a. 156-339) and SH2 (a.a. 488-579), that bind phosphotyrosine-containing sequences, as well as a central region (CH1; a.a. 340-487) that contains critical tyrosine phosphorylation sites. The phosphotyrosine-binding (PTB) domain is also referred to as phosphotyrosine-interaction domain or PID.
