Descrição detalhada : Sphingosine kinase 2 (Sphk2), along with its sister enzyme Sphk1, is an important kinase needed to form the important intra and extracellular signaling lipid mediator and neuroprotective factor SPP/S1P (sphingosine 1-phosphate). Sphk2 is important in blood vessel development, brain development, as well as an anti-apoptotic factor, and it mediates many proliferation mediated responses such as those surrounding insulin and its mitogenic ability. Sphk2 is localized to the lysosome membrane primarily but can also be found in the cytoplasm. Interestingly, recent research indicates that ApoE regulates the secretion Sphingosine 1-phosphate in the brain, and SPP/S1P in turn positively regulates glutamate secretion and synaptic strength in hippocampal neurons, thus Sphk1/2 plays an important role in synaptic plasticity and neuronal activity connectivity in the brain as well.
