Descrição detalhada : Zinc finger MYND domain-containing protein 11 (UniProt Q8R5C8) is encoded by the Zmynd11 gene (Gene ID 66505) in murine species. ZMYND11/BS69 functions as a co-repressor of adenovirus E1A and cellular transcription factors including the c-Myb and ETS-2 oncoproteins. ZMYND11 contains several histone reader motifs, including a plant homeodomain (PHD), a bromodomain, and a PWWP domain. Lys36-trimethylated histone H3 (H3K36me3) is deposited onto the nucleosomes in the transcribed regions after RNA polymerase II elongation, and ZMYND11 is found to specifically recognize H3K36me3 and regulate RNA polymerase II elongation. Binding study showed that the combined PHD-bromo-PWWP (PBP) module mediates ZMYND11 recognition of H3K36me3 and PWWP domain alone displays little to no affinity toward H3K36me, H3K36me2, or unmethylated H3K36. ZMYND11 is a tumor suppressor that functions as an unconventional transcription co-repressor by modulating RNA polymerase II at the elongation stage. Low expression levels of ZMYND11 in breast cancer patients correlate with poor prognosis. Consistently, ZMYND11 overexpression suppresses cancer cell growth in vitro and tumor formation in mice in vivo.
