Descrição detalhada : PMEF-NL-P1 are low passage neomycin resistant Primary Mouse Embryo Fibroblasts (PMEFs) isolated from FVB mouse embryos at day 13. The cells have NOT been mitotically arrested. PMEFs are the traditional feeder cells used to support the growth of undifferentiated mouse and human pluripotent stem cell cultures. Additionally, low passage primary cultures of fibroblasts have a high proliferative capacity and can be used for reprogramming. It is known that cellular reprogramming efficiency declines with each passage, presumably due to lowered proliferative capacity of PMEFs at later passages. These untreated PMEF cells are high quality, low passage (P1) mouse fibroblasts that have been successfully reprogrammed into iPS cells with high efficiency. They can be used to study the mechanisms of reprogramming or be further expanded and treated with gamma-irradiation or mitomycin-C to make growth arrested feeder cells. Many embryonic stem cell culture protocols necessitate the use of primary mouse embryo fibroblast (PMEF) cells. In these protocols, ES cells are typically cultured on a monolayer of PMEF feeder cells. Feeder cells perform two important roles in stem cell culture: they secrete several important growth factors into the medium, which help maintain pluripotency, and they provide a cellular matrix for ES cells to grow.
