Immune checkpoint molecules have been found on the cell surface of immune cells and tumor cells as activating receptors or inhibitory receptors. Immune checkpoint inhibitors, such as blocking antibodies to cytotoxic T lymphocyte-associated antigen-4 (CTLA-4), programmed cell death protein-1 (PD-1) and programmed cell death ligand 1 (PD-L1), have demonstrated significant efficacy in the treatment of a variety of cancers. Recent studies have also shown many immune checkpoint proteins can undergo alternative splicing or proteolytic cleavage to produce soluble checkpoint proteins.
